Classic Car News: Vintage Means Value | Automotive News Magazine

Paying out for that vintage car of your dreams may leave your pockets feeling light, but in the long run you may find you have more spare cash than you thought. Modern cars depreciate in value every year, however antique or classic cars not only hold their value but also increase in value over time.

?Lots of people like new cars,? says Tim Schofield, motoring expert at auctioneers and valuers, Bonhams, ?but as soon as you drive these out of the showroom, they depreciate like a stone?. The Telegraph online reports that ploughing money into a piece of ?automotive art?, also known as a classic car, could be the ideal investment.

Finding that perfect vintage vehicle can take time, with each model looking, feeling, smelling and moving differently ? with its very own personal nostalgic value. According to the Telegraph.co.uk, ?a classic car should be a worthwhile investment thanks to the unparalleled joy and satisfaction it?s capable of delivering?.

The Telegraph online reports that some wise investments could be a 1996-99 Boxter, which can be picked up for around ?6,000 or the 1989-94 911s valued at around ?8,000. If you have the cash to splash, another investment could be a mint condition Aston Martin DB4 GT from 1962-63, which is worth a staggering million (around ?680,000). However a ?dog rough 1969-73 DBS V8 is available today for ?5,000-?10,000, while an unkempt 1994-99 DB7 is a shade under ?15,000?.The joy of owning a classic car that may not be in the best condition is that you can restore it to its original beauty or keep it preserved in its current, somewhat aged state.

Fine craftsmanship, seldom seen and utterly beautiful ? it?s not difficult to see the appeal in classic cars. If you have more money and sense, then you could choose a classic car that is worth its own weight in gold, such as the Aston Martin DB5 (the car driven by James Bond in the Sean Connery era), or the Ferrari Dino and Lamborghini Miura.

According to AssociatedContent.com, the classic car trend seems to suggest that ?classics may be one of those things that won?t get crushed under the weight of the economy?, having made it through many an economic breakdown and still coming out on top.

When taking on the venture of restoring one of the legendary metal masterpieces of the past yourself, you should make sure you don?t get caught with your pants down so to speak ? the more you know, the more you can do to restore the vehicle to its original beauty.

Tim Schofield, of Bonhams believes that ?generally speaking, if a car caused a sensation when it was first launched, then it?s always going to be collectable in years to come?.

When to Refinance a Mortgage | Rai Chertkoff Real Estate Guides

Mortgage Refinance

Most people know just enough about refinancing to be cautious. Here is some information that can help the process along. Going through with refinancing without knowing the basics can lead to disaster. Don?t make a huge mistake by rushing into refinancing without knowing what you are doing or how you will benefit.

There are many homeowners who are thinking of refinancing in order to help reduce their monthly cost of their current mortgage loan. Throughout this article we will go over how and when to refinance. These recent days have brought interests rates to about 6%. Those homeowners who purchased their homes when rates were higher can benefit a great deal from the lower interest rates if they were to refinance.

When to Refinance

Refinancing worked a bit differently in the past. It was said that one should wait to refinance their mortgage until they can lower their rate by at least 2%.

However in today?s world it is recommended to refinance whenever the rates drop lower than your current rate.

Two reasons why this no longer applies.

Biggest reason this no longer applies is that the rates back in the 1980s were much lower than they are today. Refinancing from 7% to 6% is equivalent to refinancing from 14% to 12%. When refinancing you must make sure that the cost of refinancing is worth the end savings.

These days there are advertisements for mortgage refinancing and whenever you notice one that offers a lower rate than you current loan look into it. It will be worth investigating if you can save a few hundred dollars a month.

Knowing when to refinance if never a for sure thing.

Are interest rates as low as they are going to get? No one knows for sure so it can be a bit intimidating but with some patience and with a little research refinancing should be a helpful tool not a harmful one. Know what you are doing and make sure those benefits outweigh the cost of refinancing. Refinancing can be the answer that you have been looking for to get stable again.

ELVIIS Volvo C30 EV charges from any standard outlet, bills the driver (hands-on)

The electric vehicle is still a fresh enough concept that the issue of redirecting the bill when filling up at a public outlet or a friend's house is of little concern. But once EVs gain more traction and the cost of electricity during that monthly dinner party starts to run upwards of 50 bucks, you'll definitely take notice. ELVIIS, a research collaboration between Ericsson and the Viktoria Institute in Sweden, may be the solution. Instead of pulling power from the grid blindly and billing the outlet's owner, the system provides driver and vehicle information to a server before charging begins, then redirects the cost for power consumed to whatever account you've registered with the vehicle, alleviating any concern about unexpected electricity bills. ELVIIS can theoretically serve other purposes, allowing you to select the least-expensive energy source, for example, or set the car to wait to charge until late-night, when rates are lower.

We jumped into the passenger seat of one of five existing ELVIIS cars -- built around an all-electric Volvo C30 -- and poked around the charging interface. The project isn't fully operational just yet, and was running in demo mode so there's no way to verify that it works as described, but it definitely appears to be solid, with Ericsson providing financial and development support. Eventually, it will use the car's GPS to identify nearby power outlets -- for now, you need to enter a specific outlet ID. There's also an Android app that allows you to monitor and reschedule charging, so if you programmed your car to be completely charged by 7AM but realized that you need to leave at 6, you can make sure you're covered without getting out of bed. But you don't need to go anywhere to see it in action -- just click on past the break.

You are here: Home / Entertainment / Theatre is a Performing Art that has Been in Existence for Many Years

The history of theatre started more than 2,500 years in the past in ancient Greek times. Theatre as a term has Greek origins, and as an art developed largely in Athens in the beginning. Theatre activities in Greece happened in huge amphitheaters and were one of three varieties, tragedy, comedy or satyr. Following was Roman theatre. For the Romans, theatre was a performance art that was well-liked and diverse. Performances incorporated street theatre, nude dancing, acrobatics and much more. Then there was the transition into Medieval Theatre, followed by the Renaissance period, Shakespeare and the 18th, 19th and 20th century?s varying interpretations of this performance art. Theatre has a lengthy history full of details telling its evolution as a performance art.

In present times we have a handful of diverse types of theatre. We have drama, musical theatre, comedy and tragedy. With the technological advances of present times, there are also diverse technical effects that go into theatre. In addition to the playwright, producer, director and actors and actresses that bring life to a performance and the designers to supply visuals, there are the effects. With design, there are costume and scene creators. Costumes are the easy element, with scenery there are lighting effects and huge scenery pieces that have to be rapidly and effortlessly movable. For the duration of scene changes, a scenery lift may be employed. These stage lifts are typically driven by hydraulics and are ready to lift total sections of a theatre stage. There is also the music that goes into a performance and other special effects, for instance pyrotechnics.

Theatre has been around for many years, but has evolved tremendously as time has progressed. While theatre productions are nevertheless a kind of entertainment and art nowadays, they have become much more advanced. In current times there are much more sophisticated special effects that can be employed along with elaborate scenery. This live art form that has been in existence for many years will only continue to improve and evolve with technological advances.

Pub owner Karen Murphy wins appeal in TV-decoder battle against Sky

Pub owner Karen Murphy wins appeal in TV-decoder battle against Sky
It's been a few months since we last heard about the legal kerfuffle between Sky and pub owner Karen Murphy. In case you've been out of the loop, it all started when the UK broadcasting giant went after Murphy for using what was deemed to be an illegal method for screening FA Premier League matches at her, or any, bar. The Greece Nova decoder, which is considered a legal bit in the privacy of your own home, was helping Murphy bypass Sky's £480 ($740) required monthly fees for bar owners and saving her over £350 ($555) in the process. Now, over $260,000 in legal fees later, Mrs. Murphy's conviction has been overturned by the relentless High Court. The ruling allows her to keep using the troubled Greek gadget to screen any EPL game without facing any troubles -- except the occasional drunken fracas. So, now you know where to go the next time you're in Portsmouth and want to catch a good ol' footy match.

Syrian troops apparently halt Homs shelling as some wounded are evacuated

BEIRUT, Lebanon ? Syrian Arab Red Crescent workers rescued a small number of wounded civilians Friday from the hard-hit Baba Amr neighborhood of the city of Homs as representatives of the International Committee of the Red Cross and the Syrian government negotiated over a ceasefire that would allow still larger evacuations.

The barrage of mortar and rocket fire that had rained down on the neighborhood came to a halt apparently for the first time in three weeks as the negotiations took place.

Two journalists wounded Wednesday in a hail of fire that killed two of their colleagues were apparently not among the seven women and children rescued Friday and taken to a local Homs hospital. But diplomatic efforts to bring the wounded journalists to safety were continuing, and an ICRC spokesman in Damascus said Red Cross officials were trying to "evacuate all persons in need of help with no exception whatsoever."

It was unclear whether that would include not just the journalists, Edith Bouvier of France and Paul Conroy of Britain, but also wounded anti-government fighters.

Homs has become the most deadly city in Syria as the government of President Bashar Assad has sought to end a rebellion that began last year. A halt to the shelling in Homs, even for a brief period, would mark a break in the government's furious assault after weeks of bloodshed. At least 120 people have been killed in the past two days in Homs, according to activists, with many bodies remaining trapped under rubble.

Saleh Dabbakeh, the ICRC spokesman in Damascus, said he could not say whether the shelling of the area had ceased during the negotiations, but he said the fact that the talks were taking place indicated that it had.

"If we are there and trying to evacuate, I don't think there would be any kind of shelling," he said.

Dabbakeh said there was "trouble and unrest in other parts of the country," but he provided no locations. "It's localized, it's in certain provinces," he said.

Bouvier and Conroy, who entered the country illegally to cover the Syrian army's onslaught against the stronghold of the rebel Free Syrian Army, appeared in a video on Thursday asking to be allowed safe passage out of Baba Amr.

Dabbakeh said Friday's evacuations were not the first since the beginning of the most recent army campaign against Baba Amr. He said that the ICRC and the Syrian Arab Red Crescent since Feb. 11 have been able to get two convoys of evacuees out of Homs and three out of the area around the western town of Zabadani.

He also said that Syrian Arab Red Crescent volunteers have been handing out humanitarian assistance and providing first aid to residents of Homs, Syria's third largest city, at nine distribution points.

"Assistance never really stopped," said Dabbakeh. "There were certain periods of time when it was difficult, sometimes very difficult."

Meanwhile, the ICRC was continuing to negotiate an agreement between the Assad regime and opposition representatives for a daily two-hour nationwide "humanitarian pause" in the fighting to allow for deliveries of food, medicines and other supplies to violence-hit towns and cities, he said.

Syrian activists said Friday that the journalists were waiting to be evacuated until there was a diplomatic presence involved.

Lenten Crafting Ideas- 10 Ways to Craft ?For Good? | Craft Test ...

Lenten Crafting Ideas- 10 Ways to Craft ?For Good?

For the Christian community, the next 40 days leading up to Easter are a sacred time- I time to reflect, grow, and recommit to God and spiritual practice. For me, Lent is a time not of deprivation, but of random joy. I choose to interpret Lent as a season in which I can spread kindness and the essence of ?Christian values? by the things that I DO. And if you know me, crafting is what I do. So why not combine the two?

Little tags and envelopes I?m sending out as RAK?s- Random Acts of Kindness.

So, I thought I?d share some of my Lenten craft ideas and practices? maybe you?ll get inspired. Most of these are Random Acts of Creative Kindness-(RAKs or RACKs)? just little things I make and send out to the unsuspecting. My hope is that the unexpected act of beauty will brighten someones day!

Random Acts of Creative Kindness:

  • Create little notes to slip to your kids, spouse, parents, friends. Tell them that they are beautiful, special, LOVED. Yes, you say it, but having it in writing is special. (See my post about Camp Cards to see what I mean.)
  • Make up a batch of cookies or candies, add a little tag that says, ?Because you?re sweet? and pass them out to crossing guards, lunch ladies, school custodians, bus drivers, postal workers, what ever?you know, those folks to improve our quality of life but we don?t usually take the time to say thank you.
  • Are you a cardmaker? Make cards to give away. Nursing homes and after-school programs regularly need cards for residents/participants to give to loved ones for special occasions.
  • Knitter? Make a hat. Or two. Or twelve. Somewhere near you, a chemo unit or a NICU (neo-natal) unit needs caps to keep those heads warm.
  • And in that vein?. use up your odds-and-ends of yarn to make striped scarves. Donate them to a men?s shelter, homeless shelter, group home, or even your closest school.
  • Are you blessed with abundance? Donate supplies. Nursing homes, hospitals, daycares (if the products are non-toxic) and even art therapists need art/craft supplies to make the lives of their clients richer. I bet you have something to spare.
  • Use up your fabric stash making lap quilts and baby quilts. Seniors in wheelchairs love pockets in theirs, too.
  • Make jewelry? Make 10 pairs of earrings, and sell them to your friends, family, co-workers for $10 each. Donate the proceeds to the charity of your choice.
  • Each one teach one. Share your skill! Teach someone to knit, crochet, embroider, sew a straight line, or master a wrapped loop. Trust me, someone out there would benefit greatly from the skills you take for granted. And moreover, someone would love the kind, nurturing attention that instruction brings.
  • Create little plushies or pillows and give them to the Red Cross or to the Child Life Department of your local hospital. Often, in times of catastrophe, kids lose their stuffed animals, blankets and ?lovies.? It?s a small way to comfort someone who might have lost everything. In my own experience, my daughter (then 2) had to be rushed to the ER for stitches. To calm her, they gave her a little pillow to hold and squeeze when she felt scared or in pain. She?s 7 now, and STILL has that pillow. It made all the difference.

?

So, there?s my list. I know you all have good ideas to share too. Please, won?t you take a minute to leave your ideas in the comments section?

Oscars 2012: Best And Worst Hosts

From Bob Hope to Chris Rock, MTV News looks back on the most memorable Academy Awards emcees.
By Kevin P. Sullivan

Anne Hathaway and James Franco at the 2011 Academy Awards

Every year, almost as important as the Best Picture winner is the host of the Academy Awards. Both grab headlines the next day, and both are sure to endure their fair share of criticism.

With almost every host having to face the black-and-white judgments of "good host or bad host," who ranks among the best and the worst? Here's our take on the Oscar hosts worth awarding and the one's that should have been snubbed.

Best
Billy Crystal
If someone hosts the show nine times, there's probably a good reason. Crystal carved out a niche for himself as the host of the big show starting in 1990 by keeping it light and throwing in a good song-and-dance routine for good measure.

Johnny Carson
The king of late night hosted the Academy Awards five times, the third most of any host. His overwhelming popularity from "The Tonight Show" carried over into his stints as host and made him one of the most consistently successful hosts. His familiarity with celebrities and experience with off-the-cuff comedy led to one of the Oscar's most memorable lines: "I see a lot of news faces, especially on the old faces."

Bob Hope
If repeat jobs as host are a measure of success, Bob Hope takes the top prize after hosting the ceremony a record 19 times. Famously calling attention to his lack of nominations, Hope's self-deprecating humor made him an Oscar legend and a rightful spot as the show's most frequent host.

Steve Martin and Alec Baldwin
As the first paired hosts since Chevy Chase, Goldie Hawn and Paul Hogan in 1987, Martin and Baldwin played off each other well enough to earn them the title of "best in recent years." Though Martin had solo hosting duties in 2001 and 2003, it wasn't until he joined forces with Baldwin that he earned his highest praise.

Jon Stewart
The host of "The Daily Show" faced a good deal of criticism after his first stint as host in 2006. He didn't shy away from controversy, calling out the liberal politics of Hollywood and Scientology, but a positive audience response brought Stewart back for a second go two years later.

Worst
David Letterman
The late-night host's name often leads the pack when recalling the worst hosts in the history of the Oscars. His "Oprah, Uma" gag and the negative reaction it got precedes most talk of how Letterman actually did. Letterman never returned to host after his one appearance in 1995.

Anne Hathaway and James Franco
The most notorious hosts in recent years, Hathaway and Franco were supposed to welcome a younger audience and make the Oscars cool again. What resulted was an odd and scattered show that was neither very funny nor cool. Many complained the Franco had phoned it in, while others found Hathaway cloying.

Chris Rock
Rock's greatest offense as host of the Academy Awards may have just been doing his normal routine, but the audience (and Sean Penn, in particular) didn't take too kindly to the comedian poking fun at some of the faces in the crowd.

Chevy Chase
Chase hosted the show a total a two times, but only once by himself. The year after his solo gig, no one hosted, which is all you need to know about that.

Jerry Lewis
It may be hard to believe with today's bloated ceremony, but in 1959, the ceremony wrapped up 20 minutes before it was scheduled to. To kill the leftover time, Lewis vamped on stage and forced some of the night's winners to continue singing and dancing on stage. NBC eventually ended the show early, but the dead, ad-libbed airtime remains an infamous part of the Oscar legacy.

Who else should make the lists of best and worst Oscar hosts? Leave your comment below!

Tecknova: Top 5 Android apps for students

Students can now use technology not only to excel in their studies but also to employ their time in a better way.

Here, The Mobile Indian has compiled five must have applications that are available on Android Market for students. All these apps are free of cost.

This application does not need any introduction. It is one of the most used reference sites in the world and is being referred to by practically everyone. If you need to quickly look up a term or want to know something in detail, just go through the app.

The Wikipedia app on Android has more than 20 million articles in 280 languages, and is the most comprehensive and widely used reference work humans have ever compiled. You can save articles to read later or offline, search articles nearby, share articles using Android's 'share' function, or even read an article in a different language.

This is one of the best educational applications available in the Android Market today. The app has different sections that include Subjects, Calculators, Translator, Math Invasion, Web Reference and Facts.

In the subjects section, the app lists Mathematics, Physics, and Chemistry. The developers are also about to add a dictionary section, which is currently inactive. Each subject has further been divided into sub sections. So if you want to know about Torque, just click on Physics and then click on Torque; or if you want to know about Statistics, go to Mathematics.

All the three subjects have been very well covered. Of course you cannot use it for comprehensive studies, but Formulas Lite, as the name suggests, is of immense help to students who want a quick revision just before a test. Since you have it on your phone, you can read it anytime, anywhere.

Besides, Formulas Lite has a Calculator, Translator, Facts and Web Reference sections. The calculator, though, is not a normal one; in fact, it provides five different types of calculators including a scientific calculator, unit converter, function derivator, binary-hex-decimal, and simple calculator.

With Translator, you can translate any English word to any of the languages listed on this app. There are no Indian languages, though; nor did it work properly when we tried. Translator perhaps works when you are connected to the internet but since it failed each time I tried, I cannot comment on this.

As the name suggests, Web Reference enables you to search for something on Wikipedia, Physics Forum, Maths Forum or Chemistry Forum. For Web Reference too you need an internet connection. The Facts section lists interesting data under various headings like celebrity, human body etc.

So all in all, Formulas Lite is a very interesting app that will prove to be helpful for students especially, enabling quick revision whenever and wherever they want.

"Evernote turns your Android device into an extension of your brain,? its developers claim and no doubt it does that with ease. With Evernote you can create notes which can be in text, picture or voice and even video, or a mix of all these. Plus, you'll never lose these Notes as they are automatically saved on the cloud, which can be accessed anytime from any computer or mobile phone using an internet connection.

So you can take notes during class and study them later. Even if you lost your phone you can retrieve notes by accessing your Evernote app using a PC, smartphone or tablet.

Merriam Webster is one of the more famous dictionaries and is referred to by many worldwide. In addition to all the definitions from Merriam-Webster's Collegiate Dictionary, this app offers voice search - to let you look up a word without having to spell it - along with synonyms, antonyms, example sentences, Word of the Day, and more. It's a great tool for reference, education, and vocabulary building.

New Health And Fitness.Org - Health Information You Can Use

If you or someone you love has been living with back pain, then you know just how annoying it can be. Sometimes, you feel great and then suddenly you turn the wrong way and have a horrible pain shooting through your body. This article will help you with just that!

Many women suffer from back pain during pregnancy. A growing baby changes your center of gravity and causes you to lean back to counteract this, causing pain in the lower back. The best remedy for this is good posture. Sit straight and keep your shoulders back. Sit in a comfortable chair and relax. Baby your back while you wait for baby!

Don?t ignore the pain. If you know a particular activity is going to exacerbate your pain, then don?t do that activity. Ignoring it will not make it go away faster. In fact, pushing through the pain will probably result in further injury, making the pain last even longer.

When it comes to back pain, ice is usually a very effective pain reliever. When applied shortly after an injury occurs or after any activity that causes pain, it can drastically help make recovery that much faster. The best combination is ice application and massage therapy used at the same time.

Put heat on any back spasms. Make a warm compress or get a heating pad and just relax. It can soothe the muscles and relax you. Stress can be a large part of back pain, so just laying down and trying to relax can end up doing wonders for you.

Many people do not think about back pain until it is too late. Most everyone is not active enough in their daily life. It is imperative that one stays active and physically fit. To get temporary relief from back pain, consider both ice and heat for at-home treatment. Ice is most effective in reducing pain from a recent injury as it helps to reduce inflammation. Meanwhile, heat is more effective at penetrating deeply to soothe more serious injuries related to chronic back pain.

You should know that the proper sleep can help you to get rid of back pain, but more important is the actually position in which you?re sleeping. Make sure that you?re not tossing and turning and make sure that your body is aligned properly while you?re sleeping. A great pillow and comfortable mattress go a long way to helping you keep back pain at bay.

Check Out How Low The VA Rates Are And How You Can Lock In ...

Many people dream of buying a home and with the low VA Rates today make it a great time to consider taking action. If you are planning on getting a home loan, you might consider getting a VA loan. But the big questions are, why choose a VA loan over a conventional loan? Who are eligible? And last, what are the unique advantages that go with the loan?

Why Choose A Veterans Loan

With VA Rates at historic lows the Veteran Administration Loan program makes it even easier for veterans to get approval for a home mortgage. VA loans are often available with no down payment, no private mortgage insurance and no closing costs. A Veteran loan gives veterans advantages that are not available in conventional home loans.

VA Loan Eligibility: Who Are Qualified?

The following are eligible for a VA loan.

1. Former members of the military on active duty during the World War II or later and who have not been dishonorably discharged.

2. Veteran soldiers who have 90 days of service during the World War II, Vietnam War or Korean Conflict.

3. Current members of the military who served in the armed forces of a government allied with US in World War II who have not been dishonorably discharged.

4. Cadets at the United States Military, Air Force or Coast Guard Academy.

5. Public Health service members

6. Midshipmen at the United States Naval Academy.

7. Midshipmen at the United States Naval Academy.

8. National Oceanic and Atmospheric Administration officers.

9. Public Health service members

Unique VA Loan Benefits and Why It Is Right For You

Below are the top 5 unique characteristics of financing a house when you compare the VA Home Purchase Rates with a Veteran Loan.

1. No Down Payment. You can finance up to 100% of your home?s value. This is perfect for veterans who have no cash on hand for down payment. The VA will lower the fee if the borrower makes a down payment of at least 5 percent. This no-down-payment loan option continue to exhibit the lowest rate of foreclosure in the market today.

2. No Out of Pocket Closing Costs. A VA loan offers a much lower closing cost compared to other conventional loans. Essentially, you are still paying for the closing costs, only you are just including them in the price of your newly purchased home.

3. No PMI Insurance. A private mortgage insurance is not required and lenders are prohibited from requiring it. One of the many reasons why a veterans loan is preferred over a conventional loan.

4. VA Loan Limit. In most US counties, the loan limit for each applicant is $417,000. However, certain high cost countries like Hawaii and Alaska have loan limits up to $625,000 per applicant.

5. VA Loan Limit of up to $417,000. Fully eligible veterans are guaranteed a loan of $104,250 simply multiply that by 4, the maximum loan amount is $417,000 up to their county limit. VA loan limits for high cost countries were raised up to $625,000 per applicant.

Nevertheless, how good this type of loan is with extremely low VA Rates is not for everyone. They are only available to veterans, active duty service members, National Guard members and etc. For a financial peace of mind, find the right lender and purchase that house of your dreams using a VA loan.

Now easier than ever you can find the best VA homeloan? Find out in 2 minutes flat by going to VA-Loan.org and shopping for the best VA Loan Today.

To understand chromosome reshuffling, look to the genome's 3-D structure

ScienceDaily (Feb. 16, 2012) ? That our chromosomes can break and reshuffle pieces of themselves is nothing new; scientists have recognized this for decades, especially in cancer cells. The rules for where chromosomes are likely to break and how the broken pieces come together are only just now starting to come into view. Researchers at Children's Hospital Boston and the Immune Disease Institute (IDI) have helped bring those rules into clearer focus by discovering that where each of the genome's thousands of genes lie within the cell's nucleus -- essentially, the genome's three-dimensional organization -- holds great influence over where broken chromosome ends rejoin. This knowledge could shed light on fundamental processes related to cancer and normal cellular functions -- for example, in immunity.

The study team, led by Frederick Alt, PhD, director of the Program in Cellular and Molecular Medicine at Children's Hospital Boston and the IDI; and Job Dekker, PhD, co-director of the Program in Systems Biology at the University of Massachusetts Medical School, reported their results online on February 16 in the journal Cell.

In cancer cells, the process of chromosome rearrangement, or translocation -- marked by stretches of DNA physically breaking and swapping -- often results in the creation of new cancer-promoting "fusion" genes. Similarly, when a na?ve B cell starts to produce antibodies for the first time, it establishes its choice of target by breaking and recombining genes for antibody diversity.

"While chromosomal breaks and translocations are fundamental to many cancers, historically we've had no approaches to systematically study how they are generated," said Alt, who is also a Howard Hughes Medical Institute investigator and the Charles A. Janeway Professor of Pediatrics and Professor of Genetics at Harvard Medical School. "About five years ago, our group set out to generate a high-throughput approach to address this important problem in cancer biology."

To accomplish this goal, the Alt lab developed high-throughput genome-wide translocation sequencing (HTGTS, which maps "hot spots" in the genome where chromosome breaks and translocations are more likely to occur) and at a level of resolution not previously thought possible. In early HTGTS studies, they found that broken chromosomes often rearrange within themselves, as opposed to sharing pieces across different chromosomes.

To probe these findings more deeply, his laboratory joined forces with Dekker's to combine HTGTS with a method called Hi-C. Developed by Dekker's group, Hi-C measures how all the sequences in the genome are organized relative to one another in three dimensions.

The combined data revealed several related but distinct principles of how genomic organization governs chromosome rearrangements. The first is based on the slight differences in how each cell organizes its genome compared to its neighbors (referred to as cellular spatial heterogeneity of genome organization). While the genome is organized in an average fashion that is largely common across all cells of a population, each individual cell harbors small deviations from that average. This latter property allows many genes to be physically close to each other in just a small subset of cells, even if they are not close to each other in the majority of cells.

The second principle involves proximity. If two broken chromosome strands lie in close proximity within the three-dimensional space of a given cell's nucleus, they are more likely to connect. This finding is of particular importance for translocations involving DNA sequences that do not break frequently, such those involved in translocations found in various non-lymphoid tumors.

The third principle applies the first two to DNA sequences that do break frequently (such as those that drive antibody gene rearrangements during B cell development). Such sequences tend to reshuffle with the same partner sequences in those subsets of cells where the partners lie physically close together, even if the partners do not within most cells. This can fuel recurrent translocations like those seen in many lymphoid tumors.

Together, the principles highlight the relationship between proximity, genomic organization, and break frequency. "Two sequences have to be broken and physically proximal to join," Alt explains. "If two sequences are together in most cells and frequently broken, they will translocate in many cells. If they are frequently together but one of them doesn't break, or if they both break frequently but always lie on opposite sides of the nucleus, the chances that they will translocate are very low or zero. However, if both sequences break very frequently and are close together in a subset of cells, they will very frequently translocate in that subset, contributing to recurrent translocations."

"Our finding that broken chromosome segments are more likely to join with other segments within the same chromosome, rather than other, more physically distant segments from other chromosomes, likely has great relevance to cancer genomes," Alt continued. "For example, cancer treatments that cause breaks may preferentially lead to intra-chromosomal rearrangements. It may also have relevance for 'chromothripsis,' a recently discovered phenomenon in many cancers in which the sequences of one chromosome become scrambled."

The new understanding of the roles of physical spatial proximity and overall three-dimensional genome structure in chromosomal translocations opens up new avenues for deciphering how the way a cell's nucleus is organized affects the genomic disarray found in cancer and other diseases characterized by chromosome reshuffling. The study also shows the power of combining two high-throughput genomic assays -- Hi-C and HGTGS -- for studying how the organizational plan within the nucleus influences fundamental biological processes.

"We feel that our findings and the application of our approaches will provide a new lens through which to view the genomes of many different types of cancer," Alt concluded.

This study was supported by the National Cancer Institute, the National Human Genome Research Institute, the Howard Hughes Medical Institute, the Leukemia and Lymphoma Society, the W.M. Keck Foundation, the Cancer Research Institute, and the German National Merit Foundation.

Information on Sciatica | New Health and Fitness

New Health And Fitness.Org - Health Information You Can Use

Sciatica is the symptoms possibly involving leg pain, maybe with tingling, numbness or weakness that travels from the lower back down by way of the sciatic nerve into the rear aspect of the leg and buttocks. Sciatica, often referred to as radiculopathy, is only what describes the symptoms. Information on sciatica from the medical profession states this is not a diagnosis. Professional examination may reveal the cause to be a herniated disc, spinal stenosis, degenerative disc disease, or spondylolisthesis, so it is recommended that one should see their doctor to identify the problem and determine the treatment.

Many people have found relief from their sciatica pain by consuming large amounts of potassium. Potassium can be found in many foods including bananas, potatoes, and oranges. It has been found that sciatica patients who consume postassium-rich foods experience less pain than those who do not eat foods rich in potassium.

Yet another natural technique to consider is chiropractic treatment, designed to adjust or manipulate your spine, thus enhancing function and reducing pain. A chiropractor may also perform some massage and stretch, to give you some relief from your lower back sciatica.

When walking it is important to always maintain proper posture. Walk upright with your head held upward. One way of teaching yourself the proper posture when walking is to get you back against a wall so that your head heels and your buttocks touch the wall and walk outwards maintaining that exact posture. When you sit it is good to remember to keep your thighs parallel with the floor and sit in an upward position and slouch back slightly. Sleeping is always one of the key ways to maintain a day free from lower back pain caused by sciatica. If you wake up pain free you are more likely to have a pain free day. The best way to sleep at night is by sleeping in fetal position with a pillow in between your knees.

A natural approach found from researching information on sciatica is simple techniques for pain management with the practice of exercise. If the pain is severe and resistant to this treatment, the next approach may involve a more structured version of this type of solution.

There?s another natural solution to sciatica problems: Sciatol. If you think prescription drugs are not working for you anymore you should try it. Sciatol is a natural supplement that helps you support and maintain a healthy spine, nervous system, and sciatic nerve. Now that you are familiar with info on sciatica problems youll know that this natural product can help you. Sciatol helps you to reduce pain, swelling, and discomfort. Sciatol also helps you prevent muscle spasm while maintaining a smooth mobility of joints. You should try it!
